In recent years the safety of probiotics has received increasing attention due to the possible transfer and spread of virulence factors (VFs) and antibiotic resistance genes (ARGs) among microorganisms. The safety of a strain of Lactobacillus plantarum named W2 was evaluated in phenotype and genotype in the present study. Its probiotic properties were also evaluated both in vivo and in vitro, including adherence properties, antibacterial properties and beneficial effects on the growth and immunity of Pacific white shrimp, Penaeus vannamei. Hemolysis tests, antibiotic resistance tests and whole genome sequence analysis showed that W2 had no significant virulence effects and did not carry high virulence factors. W2 was found to be sensitive to chloramphenicol, clindamycin, gentamicin, kanamycin and tetracycline, and to be resistant to ampicillin and erythromycin. Most ARGs have no transfer risk and a few have transfer risk but no significant enrichment in human-associated environments. The autoaggregation of W2 was 82.6% and the hydrophobicity was 81.0%. Coaggregation rate with Vibrio parahaemolyticus (24.9%) was significantly higher than Vibrio's autoaggregation rate (17.8%). This suggested that W2 had adhesion potential to mucosal/intestinal surfaces and was able to attenuate the adherence of V. parahaemolyticus. In addition, several adhesion-related protein genes, including 1 S-layer protein, 1 collagen-binding protein and 9 mucus-binding proteins were identified in the W2 genome. W2 had efficiently antagonistic activity against 7 aquatic pathogenic strains. Antagonistic components analysis indicated that active antibacterial substances might be organic acids. W2 can significantly promote the growth of shrimp when supplemented with 1 × 1010 cfu/kg live cells. Levels of 7 serological immune indicators and expression levels of 12 hepatopancreatic immune-related genes were up-regulated, and the mortality of shrimp exposed to V. parahaemolyticus was significantly reduced. Based on the above, L. plantarum W2 can be applied safely as a potential probiotic to enhance the growth performance, immunity capacity and disease resistance of P. vannamei.

Particle surface morphology is an important factor influencing sand structure and mechanical properties. In this study, the effect of sand particle sphericity on sand direct shear performance is investigated by using the discrete element method (DEM). Two ways are adapted to simulate different approaching methods from round particles to irregular sand. The macroresponse shows that irregular sand has a higher shear strength at lower normal stress than round particles. The shape of the particle has less influence on shear strength at higher normal stress. The irregular shape of sand leads to an increase in the shear band proportion. However, the shear band proportion is not related to the sphericity. Under all conditions, particles within the shear band have a larger average rotation angle than those outside the shear band. When the particle shape approaches round (regardless of the round particle proportion and particle shape), the average rotation angle of particles within and without shear bands increase, while the coordinate number and contact anisotropy decrease.

The utilizing of traditional chemical stabilizers could improve soil engineering properties but also results in brittle behavior and causes environmental problems. This study investigates the feasibility of the combined utilization of an ecofriendly biopolymer and fiber inclusions as an alternative to traditional cement for reinforcing soft soil. A series of unconfined compression tests were conducted to examine the combined effect of the biopolymer and fibers on the stress-strain characteristics, strength improvement, failure pattern, and reinforcement mechanism of soft soil. The results show that the biopolymer associated with fibers has an unconfined compressive strength similar to that of fiber-reinforced soil. However, it then increases with different curing times and conditions, which can be up to 1.5 MPa-2.5 MPa. The combined effect of fibers and the biopolymer is not simply equivalent to the sum of the effects of each individual material. The fiber shows its role instantly after being mixed into soil, whereas the effect of biopolymer gradually appears with sample curing time. The biopolymer plays a dominant role in increasing the peak unconfined compressive strength and brittleness of soil, while the amount of fiber is crucial for reducing soil brittleness and increasing ductility. It is shown that the biopolymer not only contributes to the particle bonding force but also facilitates the reinforcement efficiency of fibers in the soil. The fibers in return assist in reducing the soil brittleness arising from biopolymer cementation and provide residual resistance after post-peak failure.

This paper presents a model for estimating the moisture of loess from an image grayscale value. A series of well-controlled air-dry tests were performed on saturated Malan loess, and the moisture content of the loess sample during the desiccation process was automatically recorded while the soil images were continually captured using a photogrammetric device equipped with a CMOS image sensor. By converting the red, green, and blue (RGB) image into a grayscale one, the relationship between the water content and grayscale value, referred to as the water content-gray value characteristic curve (WGCC), was obtained; the impacts of dry density, particle size distribution, and illuminance on WGCC were investigated. It is shown that the grayscale value increases as the water content decreases; based on the rate of increase of grayscale value, the WGCC can be segmented into three stages: slow-rise, rapid-rise, and asymptotically stable stages. The influences that dry density and particle size distribution have on WGCC are dependent on light reflection and transmission, and this dependence is closely related to soil water types and their relative proportion. Besides, the WGCC for a given soil sample is unique if normalized with illuminance. The mechanism behind the three stages of WGCC is discussed in terms of visible light reflection. A mathematical model was proposed to describe WGCC, and the physical meaning of the model parameters was interpreted. The proposed model is validated independently using another six different types of loess samples and is shown to match well the experimental data. The results of this study can provide a reference for the development of non-contact soil moisture monitoring methods as well as relevant sensors and instruments.

ETHNOPHARMACOLOGICAL RELEVANCE: Ma-xing-shi-gan-tang (MXSGT), which is documented in the Treatise on Febrile Diseases and is a therapeutic drug, is a well-known classic prescription in China and has been widely studied. Previous studies have shown that MXSGT has various pharmacological activities, including anti-influenza virus activity, and ameliorates microvascular hyperpermeability and inflammatory reactions. However, no study has reported the effect of MXSGT in the treatment of bacterial pneumonia. AIM OF THE STUDY: In this study, the potential inhibition of MXSGT against the virulence of S. pneumoniae by targeting PLY was investigated. MATERIALS AND METHODS: First, HPLC analysis was used to determine the main components of MXSGT. Then PLY protein was constructed and used for hemolysis assay and western blot to test the ability of MXSGT to inhibit PLY activity, production and widowed characteristics. The growth curve of S. pneumoniae was drawled with or without MXSGT treatment. In addition, the inhibition of MXSGT against PLY-mediated A549 cell death was examined by cytotoxicity assay. Finally, the mouse experiment was used to verify the effect of MXSGT on mouse lungs. RESULTS: This work has discovered that MXSGT, a TCM prescription, is an effective inhibitor of PLY, an important virulence factor that is essential for S. pneumoniae pathogenicity. MXSGT inhibits the oligomerization of PLY without affecting S. pneumoniae growth and PLY production. In addition, experimental MXSGT treatment was effective against S. pneumoniae infection both in vitro and in vivo. CONCLUSION: These findings directly demonstrate the potential mechanism of the Chinese herbal formula MXSGT in the treatment of pneumococcal disease and provide additional evidence for promotion of the wide use of MXSGT in the clinic.

Stabilization/solidification (S/S) has been successfully employed in many superfund sites contaminated with organic materials. However, this method's long-term effectiveness has not been fully evaluated and the increase in soil volume following treatment is unfavorable to follow-up disposal. The present study developed a novel method for the S/S of PAHs-contaminated soil with the facilitation of sulfonated oil (SO). Adding SO significantly improved the unconfined compressive strength (UCS) values of Portland cement and activated carbon (PC-AC) treated soil samples, and the UCS values of the soil sample treated with 0.02% of SO were up to 2.3 times higher than without SO addition. When the soil was treated with PC-AC-SO, the PAHs leaching concentrations were 14%-25% of that in leachates of the control soil, and high molecular weight PAHs including benzo(a)pyrene were rarely leached. Freeze/thaw durability tests reveal that the leachability of PAHs was not influenced by freeze-thaw cycles. The UCS values of PC-AC-SO treated soil samples were 2.2-3.4 times greater than those of PC-AC treated soil samples after 12 freeze-thaw cycles. The PC-AC-SO treated soils resist disintegration better when compared to the PC-AC treated soils. The SEM micrographs reveal that the soils' compactness was significantly improved when treated with SO.
